I have been using wireless activation for years. What is the difference between wireless activation and desktop manager activation? How reliable is desktop manager activation?

No difference really.

You'll find that activating over the desktop manager is usually faster than over wireless. Wireless is dependant on the mobile network signal.

If you need to re-activate a BB, do you need to first wipe everything or not?

There is a difference. Usually, if you do wired, you don't have to involve an admin or have your BES password reset. If the DM is connected to the Messaging System (i.e. in the DM, in shows Server: Online), then you just connect up and it activates.

Wireless activation has always required me to call a BES admin first and get them to reset my BES password.

No, you don't need to wipe before you re-activate. In fact, if you want to have all your old messages, etc, you should Restore them from a backup BEFORE you activate.
