I spoke to tech support of Amzer and they suggested for sync port validation which worked. I am sharing for all who often face problem like this
1 - Start the BlackBerry Desktop Manager on your computer.
2 - Connect your phone to your PC using the data cable you got with cradle. Ensure they are no loose connections.
3 - On the menu bar, click on Options > Connection Settings
4 - From the drop down beside "Connection", select <USB> and click Detect. This will automatically detect the port on which you have connected your phone
5 - Your BlackBerry Desktop Manager should automatically detect the port. Click OK
