Does your company have a BlackBerry Enterprise Server? If so, they may have blocked RIM's IP addresses so you can't set it up unless you use the server to do it (and go thru your IT dept for it).
Since you said you had it working once and could receive but not send, I'm thinking the above scenario is most likely.
No longer a BES Admin, but it was fun while it lasted!
